    Effect of Urea and Distillers Inclusion in Dry- Rolled Corn Based Diets on Heifer Performance and Carcass Characteristics

    Crossbred heifers (n=96, BW = 810 ± 20) were utilized to evaluate the effects of increasing wet distillers grains plus solubles and urea inclusion in a dry rolled corn based finishing diet on performance and carcass characteristics. Heifers were individually fed using a calan gate system with a 2 × 2 factorial arrangement of treatments. Factors included distillers inclusion at either 10 or 20% of diet DM and urea inclusion at either 0.2 or 1.4% of diet DM. Th ere was no difference for final body weight, average daily gain, and feed conversion on a live or carcass adjusted basis for either urea or distillers inclusion in the diet. Dry matter intake was reduced with increased urea inclusion; however, distillers inclusion did not influence intake. Added distillers and urea in the diet had minimal impact on performance suggesting supplemental urea in a dry rolled corn based finishing diets is of minimal benefit when feeding at least 10% distillers grains

    Expression of Human Microsomal Epoxide Hydrolase in Saccharomyces cerevisiae Reveals a Functional Role in Aflatoxin B1 Detoxification

    The metabolism and genotoxicity of the carcinogenic mycotoxin, aflatoxin B1 (AFB), was studied in the lower eukaryotic yeast Saccharomyces cerevisiae. Recombinant strains of yeast were engineered to express human cDNAs for CYP1A1, CYP1A2, and microsomal epoxide hydrolase (mEH). Coexpression of mEH with CYP1A1 or CYP1A2 resulted in significant decreases in measurements of AFB genotoxicity. In cells expressing CYP1A2 and mEH, the level of AFB-DNA adducts was decreased by 50% relative to cells expressing CYP1A2 alone. Mitotic recombination, as assayed by gene conversion at thetrp5locus, was diminished by 50% or greater in cells coexpressing mEH and CYP1A2 compared to CYP1A2 alone. The mutagenicity of AFB in the Ames assay was also decreased by approximately 50% when AFB was incubated with microsomes containing CYP1A1 or CYP1A2 and mEH versus CYP1A1 or CYP1A2 alone. The biotransformation of AFB by CYPs is known to involve the generation of a reactive epoxide intermediate, AFB-8,9-epoxide, but previous direct biochemical and kinetic studies have failed to demonstrate any functional role for mEH in AFB detoxification.     Finishing the euchromatic sequence of the human genome

    The sequence of the human genome encodes the genetic instructions for human physiology, as well as rich information about human evolution. In 2001, the International Human Genome Sequencing Consortium reported a draft sequence of the euchromatic portion of the human genome. Since then, the international collaboration has worked to convert this draft into a genome sequence with high accuracy and nearly complete coverage. Here, we report the result of this finishing process. The current genome sequence (Build 35) contains 2.85 billion nucleotides interrupted by only 341 gaps. It covers ∼99% of the euchromatic genome and is accurate to an error rate of ∼1 event per 100,000 bases. Many of the remaining euchromatic gaps are associated with segmental duplications and will require focused work with new methods. The near-complete sequence, the first for a vertebrate, greatly improves the precision of biological analyses of the human genome including studies of gene number, birth and death. Notably, the human enome seems to encode only 20,000-25,000 protein-coding genes. The genome sequence reported here should serve as a firm foundation for biomedical research in the decades ahead

    Influence of host genetics in shaping the rumen bacterial community in beef cattle

    In light of recent host-microbial association studies, a consensus is evolving that species composition of the gastrointestinal microbiota is a polygenic trait governed by interactions between host genetic factors and the environment. Here, we investigated the effect of host genetic factors in shaping the bacterial species composition in the rumen by performing a genome-wide association study. Using a common set of 61,974 single-nucleotide polymorphisms found in cattle genomes (n = 586) and corresponding rumen bacterial community composition, we identified operational taxonomic units (OTUs), Families and Phyla with high heritability. The top associations (1-Mb windows) were located on 7 chromosomes. These regions were associated with the rumen microbiota in multiple ways; some (chromosome 19; position 3.0–4.0 Mb) are associated with closely related taxa (Prevotellaceae, Paraprevotellaceae, and RF16), some (chromosome 27; position 3.0–4.0 Mb) are associated with distantly related taxa (Prevotellaceae, Fibrobacteraceae, RF16, RFP12, S24-7, Lentisphaerae, and Tenericutes) and others (chromosome 23; position 0.0–1.0) associated with both related and unrelated taxa. The annotated genes associated with identified genomic regions suggest the associations observed are directed toward selective absorption of volatile fatty acids from the rumen to increase energy availability to the host.     Dried distillers grains (DDGS) were fed to weanling steer calves grazing nonirrigated corn residue to determine daily gain response and residue intake response to increasing levels of DDGS (from 1.5 to 6.5 lb/day in 1 lb increments). The DDGS was fed individually using Calan electronic gates. Daily gain increased from 0.9 (1.5 lb DDGS) to 1.8 (6.5 lb DDGS) lb/day . Forage intake decreased from 11.3 (1.5 lb DDGS) to 8.3 (6.5 lb DDGS) lb/day . Results provide information for selecting a DDGS supplementation level to achieve a target gain

    Mineral Composition of Beef Cattle Carcasses

    Mineral retention was measured in 76 beef steers. Cattle were grown at different rates of gain and then finished on a common diet. Calcium and P retention were not affected by treatment and were similar between the growing and finishing periods averaging 4.2 g P and 10.8 g Ca /100 g protein gain across both experiments. As ADG during the growing period was decreased, K, Mg, and S mineral retention during the finishing period were increased. Expressing mineral retention as g/100 g protein gain reducedvariation due to animal size and ADG and suggests that current NRC predictions are accurate
